Single and Multiplex Assays
ELISA (enzyme-linked immunosorbent assay), also called enzyme immunoassay (EIA), is a microplate-based assay used to detect and quantify peptides, proteins, antibodies and hormones. It relies on antibodies to detect a target antigen using highly specific antibody-antigen interactions. In an ELISA, an antigen must be immobilized on a solid surface (96- or 384-wells plate) by direct binding or via a capture antibody. The immobilization of the antigen facilitates the separation of the antigen from the rest of the components in the samples.

This MSD technology is based on the electrochemiluminescent (ECL) labels that generate light when stimulated by electricity in the appropriate chemical environment. The bottom of the well contains carbon electrodes. The binding interactions between the molecules present at the bottom of the plate and the detection reagent (SULFO-TAG or Ruthenium [II] tris-bipyridine) bring the ECL label close to the carbon electrode. Upon application of an electrical current, the labels produce light at 620 nm, which is detected by the plate reader. The assays can be multiplexed by using multi-spot plates whereby up to ten spots per well can be conjugated to different analytes. This technology provides a very efficient way of analyzing samples from subjects vaccinated with multi-valent vaccines, greatly decreasing the processing time in the lab.

AlphaLISA is the only truly homogeneous ELISA alternative allowing all-in-one-well immunoassays with no wash steps. The technology is based on the luminescent oxygen channeling between donor and acceptor beads. Basically, binding of molecules conjugated on the respective beads leads to an energy transfer from one bead to the other. The intensity of the generated signal is proportional to the amount of bound proteins. The assays can be performed in duplex or triplex format using acceptor beads emitting light at different wavelengths. The signals are read on the EnVision® luminescence plate reader from PerkinElmer.
